(a) Downregulation of Th1 cytokine production in SS model mice treated with Clik60. Culture supernatants from anti-CD3 mAb–stimulated splenic T cells from Clik60-treated model mice (8 weeks old) contained a high level of IL-4, but low levels of IL-2 and IFN-γ, as measured by ELISA (*P < 0.005, **P < 0.001, Student’s t test). (b) No significant cytokine production in culture supernatants from anti-CD3 mAb–stimulated splenic T cells from non-Tx NFS/sld mice (8 weeks old). (c) Production of serum autoantibodies to 120-kDa α-fodrin was diminshed in two mice (no. 1 and no. 2) treated with Clik60, as shown by immunoblotting. (d) A dilution curve illustrates that each case is capable of reacting with antigen. The same dilution of sera was examined in all cases by ELISA. OD492, optical density of 492 nm.
