Endothelial NOS (eNOS), and its product NO, are vital components of the control of
vasomotor function and cardiovascular homeostasis. In the present review, we will take a
deep dive into eNOS enzymology, function and mechanisms regulating endothelial NO. The

In the present review, we will take a deep dive into eNOS enzymology, function and mechanisms regulating endothelial NO. The mechanisms regulating eNOS and NO synthesis discussed here include alterations to transcriptional, post‐translational modifications and protein–protein regulations. Also, we will discuss the phenotypes associated with various eNOS mutants and the consequences of a disrupted eNOS/NO cascade, highlighting the importance of eNOS function and vascular homeostasis.
