Peptide-induced upregulation of MHC class I on TAP-deficient T2 cells is used to monitor
peptide binding to class I molecules. We present a temperature-dependent modification of
the T2 assay which increases the sensitivity of peptide binding to MHC class I with several
orders of magnitude.
