Cytokines and hypoxia associated with worsening of diabetic retinopathy increase PSF expression. (a) Confluent cultures of RECs were exposed to TGF-β (1 ng/mL), VEGF (25 ng/mL), bFGF (10 ng/mL), or hypoxia (0.5% O2, 5% CO2, 94.5% N2) for 24 hours before evaluation of the media for PSF protein by Western blot analysis. (b) Nearly confluent cultures of RECs were exposed to hypoxic conditions (0.5% O2, 5% CO2, 94.5% N2) for the period of time indicated in the figure before isolation of RNA and Northern blot analysis.
