Figure 4sLRP-α selectively regulates TNF-α–initiated cell signaling. Top: Protein extracts from Schwann cells in culture were subjected to SDS-PAGE and immunoblot analysis to detect phosphorylated ERK1/2 (P-ERK1/2) and total ERK1/2 (T-ERK1/2). The cells were first treated with sLRP-α (50 nM) or vehicle for 10 min prior to adding PDGF-BB (1.0 nM) for 10 min. Control cells were not treated with PDGF. Middle: Schwann cells were pretreated with sLRP-α (50 nM) or vehicle for 10 min and then with 1.0 nM erythropoietin (Epo) for 10 min. Immunoblot analysis was performed to detect P-JAK2 and β-actin.
